Super-Enhancer Condensates: Phase Separation & BET Inhibition (2D)

Interactive 2D particle simulation of biomolecular condensate formation at super-enhancers: multivalent coactivator molecules undergo liquid-liquid phase separation on canvas via real overdamped Langevin dynamics, and a BET-inhibitor dose slider dissolves the droplet exactly as JQ1 does in real cells.

Transcriptional coactivators cluster into liquid-like biomolecular condensates at super-enhancers, concentrating the transcription machinery on the genes that define cell identity. This 2D canvas simulator models a population of multivalent coactivator molecules diffusing under overdamped Langevin dynamics with a short-range sticker-and-spacer attraction: raise the valency or concentration and the dispersed gas of particles nucleates into a dominant droplet; dial in a BET-inhibitor (JQ1) dose and watch the same droplet dissolve as the drug competitively blocks the bromodomain-acetyl-lysine contacts holding it together. Live readouts track the largest condensate's share of all molecules, the number of distinct droplets, and the effective interaction-to-thermal-energy ratio that governs the phase transition.

⚙ Under the hood

Watch multivalent transcriptional coactivators undergo liquid-liquid phase separation into a super-enhancer condensate, then dial in a BET-inhibitor dose to see the same droplet dissolve as it competitively blocks the bromodomain contacts holding it together.
