A physiologically-based pharmacokinetic (PBPK) model of a single IV bolus dose as it circulates through the body: blood is pumped to the liver, kidney, fat and the rest of the body at each organ's real fraction of cardiac output, each tissue equilibrates with blood through a drug-specific partition coefficient, and hepatic and renal clearance permanently remove drug from the liver and kidney compartments. Adjust the dose, the drug's lipophilicity, and CYP450/renal clearance to see how a hydrophilic drug stays confined to blood while a lipophilic one is sequestered — and slowly released — by fat, with every organ's live concentration tracked numerically alongside the flowing 3D circulation.
