Exon-Skipping ASO — Frame-Shift Phase Diagram

The 2D counterpart to the 3D ribosome scene: the same exon-51-skipping antisense-oligonucleotide arithmetic for Duchenne muscular dystrophy, redrawn as a dose-response curve, a mod-3 reading-frame phase clock, a codon piano-roll, and a stochastic population grid.

This is the 2D counterpart to the 3D ribosome scene, driven by the identical reading-frame arithmetic behind splice-switching antisense-oligonucleotide therapy for Duchenne muscular dystrophy (the eteplirsen/exon-51 drug class). Instead of a spatial ribosome translating a 3D mRNA track, this view plots the same Σ(bases removed) mod 3 formula as a dose-response sigmoid you can read a probability off of, a pair of mod-3 phase clocks whose needle angle is the frame shift itself, a piano-roll codon track swept by a read head, and an independently-resampled Monte-Carlo grid showing what fraction of a transcript population is rescued at the current ASO dose.

⚙ Under the hood

The 2D counterpart to the 3D ribosome scene: pick a Duchenne muscular dystrophy patient's dystrophin deletion genotype and dose an exon-51-skipping antisense oligonucleotide, but read the reading-frame arithmetic off a dose-response sigmoid, a pair of mod-3 phase clocks, a codon piano-roll track, and a resampling Monte-Carlo population grid instead of a spatial ribosome animation.
