Hypoimmunogenic Islet Transplant Simulator — 2D

Interactive 2D simulator of a gene-edited, hypoimmune iPSC-derived pancreatic islet graft: toggle HLA class I/II knockout, CD47 and PD-L1 overexpression, drag to pan and scroll to zoom the liver-sinusoid view, and watch T cells and NK cells decide whether the graft survives without immunosuppression.

Type 1 diabetes cell therapy (as in Vertex's VX-880 iPSC-derived islet programme) still needs lifelong immunosuppression because allogeneic islets carry donor HLA molecules that a recipient's T cells recognise as foreign. Gene-editing the graft instead — knocking out HLA class I and II, then adding CD47 and PD-L1 as immune checkpoints — aims to make the cells invisible to rejection without suppressing the whole immune system. This 2D simulator drops an islet-cell cluster into a liver-sinusoid vessel alongside patrolling T cells and NK cells, runs a daily stochastic rejection model driven by whichever edits and immunosuppression level you choose, and tracks graft survival and insulin output over two simulated weeks of engraftment — showing why HLA-I knockout alone backfires (NK "missing-self" attack) unless CD47 is added alongside it. Pan and zoom the pannable arena to inspect individual cell fates up close.
