Hydrogen-deuterium exchange mass spectrometry probes a biologic's higher-order structure (HOS) by soaking it in heavy water and measuring how fast backbone amide hydrogens swap for deuterium — buried, hydrogen-bonded regions exchange slowly while flexible surface loops exchange almost instantly.
