CRISPRi/CRISPRa Positional Efficacy Simulator (2D)

Drag a dCas9-KRAB (CRISPRi) or dCas9-VPR (CRISPRa) effector along a 2D gene locus diagram and watch a live efficacy-vs-position curve, transcription rate and RNA Pol II traffic respond to distance from the transcription start site.

Epigenomic editors don't cut DNA — they occupy it. This 2D companion places a dCas9 fusion (KRAB for CRISPR interference, or VPR for CRISPR activation) at a chosen offset from a gene's transcription start site and computes a real positional efficacy curve for how strongly that effector controls transcription from there, based on the CRISPRi/CRISPRa screening literature. Drag the effector along the gene diagram or use the slider to explore each effector's bell-shaped sweet spot, add guide RNAs to see diminishing-returns multiplexing, and watch the live efficacy-vs-position chart, RNA Polymerase II traffic and transcription rate respond in real time.

⚙ Under the hood

Move a dCas9-KRAB (CRISPRi) or dCas9-VPR (CRISPRa) effector along a gene locus and watch a real positional efficacy curve, RNA Pol II traffic, and chromatin openness respond as its distance from the transcription start site changes.
