Saturation genome editing tests every possible point mutation across a gene exon in a single pooled experiment, sequencing cell populations before and after selection to score how each variant affects fitness — the same approach Findlay et al. used to functionally classify thousands of BRCA1 variants before they were ever encountered in the clinic.
