Thermal Proteome Profiling: 2D Melt-Curve Dashboard
Interactive 2D thermal proteome profiling (CETSA/TPP) dashboard: heat-challenge a target protein population and an off-target control on a canvas rendering of the centrifuge tube, add a ligand at variable concentration and affinity, and watch soluble-fraction melt curves and the apparent Tm shift plotted live alongside a binding isotherm.
Thermal proteome profiling heats a protein population, removes whatever aggregates by centrifugation, and reads out the soluble fraction left behind — the same read-out mass spectrometry captures at proteome scale. This 2D canvas dashboard renders a target protein population and an unrelated off-target control side by side as schematic centrifuge tubes, each of the 30 copies per group carrying its own randomly offset melting threshold so the group denatures along a real curve rather than all at once. Two live charts underneath turn that per-particle simulation into the actual textbook read-outs: a soluble-fraction-vs-temperature melt curve (the exact cumulative-Gaussian formula the particles obey) and a ΔTm-vs-concentration binding isotherm. Add a ligand at a chosen concentration and binding affinity and watch the target's apparent melting temperature shift upward, while the control — which the drug does not bind — stays put, exactly the specificity signature that identifies a genuine drug target in a real TPP/CETSA screen.
