Cartilage Scaffold Oxygen Diffusion & GAG Synthesis (2D)
2D cartilage-scaffold bioreactor lab: a finite-difference O2 reaction-diffusion solver across scaffold depth drives Michaelis-Menten chondrocyte oxygen consumption, a strain-dependent GAG synthesis rate, and a live hypoxic-core readout.
This 2D companion pulls the diffusion-limit problem out of the 3D MACI pipeline narrative and solves it directly: a finite-difference reaction-diffusion field simulates oxygen penetrating a cartilage scaffold from its media-facing surface while chondrocytes consume it via Michaelis-Menten kinetics, so thicker or more densely seeded constructs develop a genuine, live-computed hypoxic and eventually necrotic core — while a strain-gated synthesis model integrates GAG (matrix) accumulation and derived construct stiffness over simulated culture days, exposing four real levers (scaffold thickness, seeding density, mechanical loading and media O2 tension) instead of the stage-narration the 3D version uses.
2D cartilage-scaffold bioreactor lab: a finite-difference O2 reaction-diffusion solver across scaffold depth (∂C/∂t = D·∂²C/∂x² − Vmax·C/(Km+C)) drives Michaelis-Menten chondrocyte oxygen consumption, with a strain-dependent bell-curve GAG synthesis rate integrated over simulated culture days into a live hypoxic-core and construct-stiffness readout.
