1. CRISPR-Cas9 Mechanism

2. CRISPR Variants & Improvements

Base editors: CBE (C→T) and ABE (A→G) without DSBs — 8th nucleotide window, 50–80% efficiency. 3. Delivery & Manufacturing

Clinical trials (2024–2026): CRISPR Therapeutics CTX001 (blood disorders), Intellia NTLA-2001 (transthyretin amyloidosis — in vivo liver editing, 87% protein reduction), Editas EDIT-101 (Leber congenital amaurosis, retinal in vivo editing). Cancer: allogeneic CAR-T with CRISPR knockout of TCR/MHC (universal donor T cells). Infectious disease: excision of HIV proviral DNA, antiviral against SARS-CoV-2 RNA (Cas13). Agriculture: non-GMO regulatory path in many countries — disease-resistant crops, improved nutrition.

5. Ethics & Regulation

Germline editing: heritable changes — He Jiankui's CCR5-edited twins (2018) widely condemned, resulted in imprisonment. International moratorium on clinical germline editing (NAS, WHO). Somatic editing: non-heritable, broadly accepted for therapeutic purposes. Equity: CRISPR therapies cost $2–3 million per patient (Casgevy) — access concerns. Gene drive: force trait through population (e.g., malaria-resistant mosquitoes) — ecological risk assessment needed. Dual use: bioweapon potential, gain-of-function concerns.
