Genetic Code Expansion: Amber Suppression Race (2D)

Watch ribosomes translate a transcript with an engineered amber (UAG) stop codon recoded to carry a non-canonical amino acid. At that codon, every ribosome races an orthogonal ncAA-charged suppressor tRNA against release factor 1 (RF1) — a real kinetic-partitioning competition used to biofabricate site-specifically labelled therapeutic proteins such as antibody–drug conjugates. Tune ncAA concentration, orthogonal tRNA/synthetase expression and RF1 activity (or switch to an RF1-attenuated chassis strain) and watch the measured full-length yield converge on the theoretical suppression-race ratio.
