SN-38 molecule UGT1A1 enzyme cluster

UGT1A1 Irinotecan Toxicity Simulator (2D)

Irinotecan is converted in the body to SN-38, a potent topoisomerase-I inhibitor that is cleared almost exclusively by the liver enzyme UGT1A1 through glucuronidation. Patients carrying reduced-function UGT1A1 alleles (most notably *28) clear SN-38 more slowly, so the same chemotherapy dose produces a larger drug exposure and a much higher risk of severe neutropenia. This 2D simulator runs the same real one-compartment pharmacokinetic model (the Bateman function) as its 3D sibling, drawing a live concentration-time curve with the AUC shaded underneath and a pannable, zoomable orbit view of SN-38 molecules circling a UGT1A1 enzyme cluster: pick a genotype, set the dose, and watch the plasma concentration curve, the cumulative AUC, and a live Emax-model neutropenia risk grade respond exactly as the pharmacogenomics literature describes.
