Microfluidic LNP Nanoprecipitation

Every mRNA vaccine dose starts as two liquid streams merging inside a microfluidic chip: lipids dissolved in ethanol, and mRNA dissolved in an acidic aqueous buffer. This simulation renders that mixing channel in 3D — an ethanol stream of ionizable-lipid and PEG-lipid particles (blue) meets an aqueous mRNA stream (orange), and the two are folded together by grooved staggered-herringbone ridges on the channel floor. Tune the total flow rate, the aqueous:organic flow-rate ratio and the lipid concentration, or switch the herringbone grooves off to fall back to slow diffusive mixing, and watch the Reynolds number, mixing completeness, resulting nanoparticle diameter, polydispersity index and estimated mRNA encapsulation efficiency respond live as particles nucleate into lipid nanoparticles at the channel outlet.
