Capillary Isoelectric Focusing of Proteins

Capillary isoelectric focusing (cIEF) separates proteins not by size but by charge: a stable pH gradient runs the length of a narrow capillary, and each protein migrates under an electric field until it reaches the point where the local pH equals its own isoelectric point (pI) — where its net charge is exactly zero and the driving force vanishes. This simulator models five real proteins spanning pI 1.0 to 10.2 (pepsin, β-lactoglobulin, myoglobin, ribonuclease A and cytochrome c) as they self-focus into sharp bands, with a live pH-dependent net-charge model, thermal diffusion opposing the focusing force, and a real-time detector-style density profile beneath the 3D capillary.
